Title: DNA FINGERPRINTING OF NATURAL POPULATION OF PUCCINIA STRIIFORMIS F. SP. TRITICI IN TIANSHUI AREA
Abstract: By employing PSR331S3 as a probe and BglⅡ as restriction endonuclease, the fingerprints of Puccinia striiformis f. sp. tritici population containing 244 isolates from Tianshui area of Gansu province were investigated. Totally 185 phenotypes of fingerprint were identified in the tested pathogen collection, the genetic polymorphism Shannon M value was 4.467, and normalized M* was 0.9106. The population M values of isolates collected from Fenghuang, Gangu and Maiji mountain within Tianshui area were 3.5720, 3.0268 and 3.4186, while their normalized M* were 0.9330, 0.8513 and 0.8610 respectively. This research revealed that the population of P. striiformis f. sp. tritici in Tianshui area was greatly rich in genetic variations. The elevation may cause the alterations of pathogen population diversity. The genetic differentiation coefficient GST value among three main locations within Tianshui area was 0.03167, which indicated that there existed a very low differentiation level among them, and also meant the exchange for the pathogen within the area was undergone very often.
