Title: Determination of Quercetin in Rutin with the Method of RP-HPLC
Abstract: [Objective] The the content of quercetin in rutin,tested by means of the method of high performance liquid chromatography(RP-HPLC),was established.[Method] The testing condition was the size of the column C18 was 250 mm × 4.6 mm,the mobile phase was water-acetonitrile-acetic acid of 1 060∶350∶45(V∶V∶V),the flow rate was 1.0 ml/min and the detection wavelength was 254 nm.[Results] Under the condition of the water-acetonitrile-acetic acid system being taken as the mobile phase,the separation of peak was good and with good type;the separation time was short and the result was accurate.Under the condition of the methanol as the solvent,the extraction time was short and the content of impurity in extract was less.There was a good linear relationship while the range of the content of quercetin was 4-12 mg/L,and the regression equation was Y=1 285X+3 645,the average recovery was 99%.The average content of quercetin in tested sample was 0.813 g/ml.[Conclusion] The method was simple,sensitive,accurate and reproducible,which could be used as an effective method in its quality and production control.
