Title: Screening of human lung cancer metastasis-associated genes
Abstract: Objective:To analyze differences in the gene expression profiling between human lung giant cell carcinoma cell strains of high metastatic 95-D and low metastatic 95-C,and to screen lung cancer metastasis-associated genes using cDNA microarray.Methods:The mRNAs were extracted from 95-D cell and 95-C cell respectively.We used genechip to obtain profiling of genes differentially expressed in the two cell lines.The expression changes of a part of genes determined in genechip were further identified by RT-PCR.Results:The analysis of genechip showed that the expression of 389 genes were changed.Among them,121 genes were up-regulated and 268 genes were down-regulated.The results of RT-PCR were coincided well with that of gene chip.Conclusions: there are significant differences in gene expression profiling between two cell lines,and the meaning of difference in expression profiling is under investigation.
