Title: Protection of Danshen on tubular cell induced by renal ischemia reperfusion injury in rats
Abstract: Objective:To investigate the protection of Danshen on tubular cell induced by renal ischemia reperfusion injury in rats.Methods:Male,Wistar rats were randomly divided into 4 groups:sham surgery group,model group(renal ischemia reperfusion injury group),medication group 1(drug given before renal ischemia reperfusion 24 hours) and medication group 2(drug given after renal ischemia reperfusion 12 hours).Renal ischemia reperfusion injury models were set up by clamping both sides of renal arteries 22 minutes.The level of serum creatimine and blood urea nitrogen was measured by colorimetry,protein expressions of renal cysteinyl aspartate-specific protease(caspase-3) were detected by western blotting.Cell apoptosis was confirmed by Terminal Deoxynucleotidyl Transferase Biotin-dUTP Nick End labeling(TUNEL) analysis.Results:Comparing the sham group and the model group,the kidney function decreases sharply(P0.05);protein expressions of caspase-3 increases remarkably(P0.05);lots of tubular cell apoptosis(P0.05).Using Danshen 24 huors before reperfusion was found remarkably improve renal function(P0.05),decrease the expressions of caspase-3(P0.05) and cell apoptosis(P0.05).And using this medication 12 hours after reperfusion,administrating Danshen did not show us any effect on renal function,cell apoptosis,and the expressions of caspase-3(P0.05).Conclusion:Before renal ischemia reperfusion,using Danshen for rats,that has the function of decompressing the cell apoptosis induced by renal ischemia reperfusion injury and protection the renal function.
