Wistar rats were randomly divided i...To investigate the expression of three isoforms of NOS in the retina before the diabetic retinopathy formation, and the correlation with retinal blood flow in rats. Wistar rats were randomly divided into control (n=10) and streptozotocin induced diabetic group(n=10). Circulatory parameters were measured using color Doppler sonography, and protein expression of eNOS, iNOS, and nNOS in rat retina were examined by immunohistochemistry. The diabetic rats were maintained for 8 weeks while diabetic retinopathy was not found. However, the circulatory parameters of central retinal artery (CRA) changed. Compared with the control group, the velocity of CRA in diabetic rats decreased significantly, but resistive index increased significantly. Pulsitive index was higher in diabetic group than in control group, but no significance was found. The expression sites of the three isoforms of NOS were the same in the rat retina of the two groups. In diabetic retina, iNOS immunoreactivity increased, eNOS and nNOS immunoreactivity maintained unchanged in comparison to that in the control retinas. [Conclusion]Decreased blood flow perfusion in the retina occurred before the formation of diabetic retinopathy, iNOS expression is upregulated in the layers of the retina and considered to be related to retina ischemia.Read More
