Title: Effect of High Glucose Environment on Cell Viability,NO,ET-1,MDA and SOD in Cultured Endothelial Cells
Abstract: Objective To detect the nitric oxide(NO),endothelin(ET),malondialdehyde(MDA)and superoxide dismutase(SOD)in cultured endothelial cells exposure to different glucose concentration,and investigate the effect of high glucose concentration ET,SOD,NO and MDA.Methods The levels of NO,MDA,ET,and SOD in the supernatants were detected by nitrate reductase test,thiobarbituric acid test,radioimmunity method and xanthine oxidase test,respectively.Results When endothelial cells were cultured in high glucose,the activities of SOD and the level of NO were significantly decreased,but the levels of ET-1 and MDA were markedly increased.Conclusion High glucose disrupts the oxidative equilibrium and decreases the vascular elasticity.
