Title: The time relationship of IL-1β,c-fos and CRF expression in the region of hippocampus of rat models of cerebral ischemia-reperfusion
Abstract: Objective To investigate the time relationship of expression of IL 1β,c fos and corticotropin releasing factor (CRF) in the region of hippocampus MCAO rats models of cerebral ischemia reperfusion Methods Cerebral ischemia reperfusion rat models were made with left middle cerebral artery occlusion (LMCAO) by a suture According to the literatures the rats were sacrificed at time points from 30 minutes to 7 days after onset of the reperfusion Sham operated rats were regarded as control groups We measured the expressions of IL 1β, c fos and CRF protein in the region of hippocampus in brain tissue of every group with immunohistochemistry Results IL 1β immunoreactive cells increased significantly in the ischemic region as early as 1h (97 4±39 3) after MCAO and continued to increase and peaked at 2h(610 67±188 4) after reperfusion, even until the 7th day (6 6±5 9) after that, the expression of IL 1β was found at very low level in all regions examined For the protein of c fos, its expression started from 2 h (162 4±24 75), and reached a peak at 4h(521 7±103 5), up to 4th day its expression disappeared nearly The expression of c fos was most intense at 4h, but became undetectable after 4d(10 4±9 7) in the hippocampus The expression of the protein of CRF started at 2 h (97 2 ±18 4 ) and peaked at 12 h (374 4±56 95) and on 7th day (396 3±50 98) Conclusions The time of the beginning and the peak of the protein expressions of IL 1β, c fos and CRF following cerebral ischemia reperfusion showed some regularity of time
