Abstract:The objective of this study was to establish a HPLC method for simultaneous determination of 4 kinds of flavonoids(quercetin-3-galactoside,quercetin-3-glucoside,quercitrin,and quercetin) in the leaves...The objective of this study was to establish a HPLC method for simultaneous determination of 4 kinds of flavonoids(quercetin-3-galactoside,quercetin-3-glucoside,quercitrin,and quercetin) in the leaves of Rhododendron pulchrum.The following system was found to be optimum for the analysis:Waters C18 column(4.6 mm×250 mm,5 μm) by gradient elution with mobile phase A(water) and B(methanol):0-24 min,31%-42% B;24-30 min,42%-50% B;30-35 min,50%-60% B;35-40 min,60% B.The detection wavelength was 356 nm,and the rate flow was 0.7 mL/min with column temperature at 30℃.Under these conditions,4 types of flavonoids could be well separated.The average recovery of 4 types of flavoniods(quercetin-3-galactoside,quercetin-3-glucoside,quercitrin,and quercetin) were 97.73%,101.71%,98.70% and 104.28%,and the relative standard deviations of the method were 2.25%,1.31%,1.82% and 2.81%(n=5),respectively.The method was accurate and sensitive,which could be used for the determination of flavoniods in the leaves of R.pulchrum.The contents of flavonoids were in the highest levels in april.Read More
