Title: Research progress of long-acting protein therapeutics
Abstract: Due to their highly target specific recognition and high affinity which could not be mimicked by smaller chemical molecules, protein therapeutics play important roles in the treatment of many critical human diseases. However, because proteins are inherent complex, instable and susceptibility to endogenous protease and circulate clearance, they usually have short in vivo half-life. This is a crucial obstacle for their more successful development. Therefore, using protein engineering techniques and modification manipulation to increase the protein's halflife in vivo is being a most active field in the new protein drug development. This review focused on this topic, and provided an overview of several current popular strategies for extension the half-life of protein therapeutics.
