Title: Effect of RNA interference PDECGF genes on expression in the t24 cell lines of bladder carcinoma
Abstract: Objective To investigate inhibition of small interfering RNA (siRNA) on PDECGF gene expression in bladder carcinoma t24 and carcinoma proliferation and invasion.Methods siRNA aiming human PDECGF gene synthesized in vitro was labeled by fluorescence and transfected t24 cells. The transfective rate of siRNA were detected by flow cytometry. The inhibitions of siRNA on PDECGF expression, cellular growth and invasive ability were determined by Western blot, MTT and Boyden cabin method.Results The PDECGF expression in siRNA transfective group obviously decreased. The inhibitive rate of cellular growth and proliferation in siRNA transfective group increased with treated time. The invasive ability in siRNA transfective group was significantly lower than that of control group and no-vector group.Conclusions siRNA of PDECGF genes can effectively down-regulate the expression of PDECGF genes and inhibit the proliferation and invasive ability of t24 cells.
