Title: Study on in Vitro Cultural System of Blastomeres Isolated from Mouse 4, 8-cell Embryos
Abstract: CZB medium was used as a base medium to study the function of taurine, glucose and porcine oviductal epithelial cells system in the culture of blastomeres isolated from mouse 4?8-cell embryos. These results showed addition of taurine in CZB did not significantly affect the blastocyst rate of blastomeres isolated from mouse 4, 8-cell embryos (29% vs 30% and 14% vs 14%) (P 0.05). Addition of glucose in CZB had a significant effect on the blastocyst rate of blastomeres isolated from mouse 4,8-cell embryos (29% vs 36% and 14% vs 19%) (P 0.05). Addition of taurine did not have a significant effect on the blastocyst rate of blastomeres isolated from mouse 4?8-cell embryos respectively(36% vs 38% and 19% vs 20%) when glucose had been added in CZB(P 0.05). The cell number of blastocysts derived from blastomeres isolated from mouse 4, 8-cell embryos were not different significantly(10.44±1.24)~(12.43±1.18) and (7.57±0.97)~(8.48±1.16) respectively (P 0.05). The blastocyst rate of co-cultured blastomeres isolated from mouse 4, 8-cell embryos were 47% and 26%, respectively; the cell number of blastocysts derived from co-cultured blastomeres isolated from mouse 4, 8-cell embryos were 17.57±1.13 and 11.43±0.92 , repectively. These were more than those of blastomeres in the controls(P 0.05).
