Title: Expression and regulation of estrogen receptor in vascular smooth muscle cells of rat
Abstract: Objective: To investigate the effects of estrogen and/or serum on the expression of estrogen receptor (ER) in vascular smooth muscle cells (VSMCs). Methods: The expression of ER mRNA in VSMCs of rats was examined by in situ hybridization (ISH) and RT PCR. The expression of ER protein was observed by immunocytochemistry (ICC) staining. The effects of estradiol (E 2) and/or newborn calf serum (NCS) on expression of ER in VSMC were compared with and without tamoxifen (TAM) (ER antagonist) treatment. Results: ER mRNA and protein were expressed in VSMC, and both E 2 and NCS could up regulate ER content in VSMC; TAM could block their effects. Conclusion: This study suggests that ER is one of the target genes of E 2 in VSMC.
