Title: Effect and impact of mailuoning on cerebral ischemia-reperfusion injury and vascular endothelial growth factor in rats
Abstract: Objective To investigate the therapeutic effect of mailuoning injection on cerebral ischemia -reperfusion injury in rats and its possible mechanisms.Methods Fifty-four SD rats were randomly divided into sham operation group, ischemia-reperfusion (IR) group and treatment group (n = 9 in each group).Functional score was performed at 6 hours after modeling.Mailuoning 1.5 mL (40 g · kg~(-1)· d~(-1)) was injected at different time points (6, 24, 48 h, and 7 d) respectively via tail veins for 7 days in the treatment group.The equal volume of isotonic saline was given at the corresponding time points in the sham operation and IR groups.The rats were scored and sampled after the end of treatment (at days 14). The cell morphology in the hippocampal CA1 region was observed with electron microscopy, and immunohis-tochemistry was used to detect vascular endothelial growth factor (VEGF) positive cells.Results ①The neurological scores in the 6, 24, 48 h, and 7 d groups were 3.2 ±0.8, 3.3 ±0.5, 3.5 ±0.8 and 4.5 ± 0.8, respectively, and they all lower than those in the IR group (5.5 ± 1.1) ; and the neurological scores within 48 h in the treatment group were lower than those in the day 7 group.There were significant differences (P0.05).②The ultrastructure of rat hippocampus in the treatment group (6, 24, 48 h, and 7 d groups) was improved more significantly than that in the IR group, and the 6h group recovered best.③The proportions of VEGF-positive cells in rat brain tissue in the treatment group (6, 24, 48 h, and 7 d groups) were 19.4 ± 1.3% , 17.6 ± 1.3% , 17.2 ± 1.5% , and 12.6 ± 1.6% , respectively, and they were all higher than 6.1 ± 1.3% in the IR group.There were statistically significant differences (P 0.01) , especially the expression level of VEGF in the 6 h group was the highest (19.4 ± 1.3%).Conclusion Mailuoning injection has better therapeutic effect on early cerebral ischemia-reperfusion injury in rats.The efficacy is better when given within 48 h, and it may play a role by increasing the expression of VEGF.
