Title: Effects of sufentanil pretreatment on hippocampus CA1 and total calcium of brain tissue in rats with cerebral ischemia-reperfusion
Abstract: Objective To investigate the effects of sufentanil pretreatment on hippocampus CA1 and total calcium of brain tissue in rats with cerebral ischemia-reperfusion.Methods Seventy male SD rats were randomly divided into five groups: sham group(group S),ischemia-reperfusion group(group I/R) and different concentrations of sufentanil pretreatment groups(group L,M,H).The rats in group I/R and sufentanil pretreatment groups received normal saline and sufentanil infusion intravenously at 0.5,1.5 and 4.5 μg/kg in the regime(3×5 min infusion with 5 min interval) before cerebral ischemia.Four-vessel occlusion was used to establish the global cerebral ischemia reperfusion model in the study groups.An acute experimental cerebral ischemia rats model induced for 15 min,24 hours after reperfusion,randomly selected six rats to observe light microscope morphological changes of the hippocampus CA1,randomly selected two of the six rats to observe electron microscopy changes of the hippocampus CA1 and the remaining eight rats to measure the total calcium in brain tissue in each group.Results Compared with group S,the hippocampus CA1 injuries,histological grades and the brain total calcium increased significantly in group I/R,L,M and H(P0.05).Compared with group I/R,the hippocampus CA1 injuries and the histological grades decreased in group M(P0.05),the brain total calcium decreased in group L,M and H(P0.05).Compared with group M,the histological grades were no significant in group L and group H,the brain total calcium increased in group L(P0.05),in group H were no significant(P0.05).The hippocampus CA1 electron microscopy: group L,M and H were less injury than group I/R in nuclear membrane,rough endoplasmic retieula and synaptic.Conclusion Sufentanil pretreatment can protect rat brain against ischemia-reperfusion injury,especially 1.5 μg/kg.
