Title: Protective effects of puerarin on cerebral ischemia-reperfusion injury in rat
Abstract: OBJECTIVE Research the protective effects of puerarin on cerebral ischemia-reperfusion injury in rats.METHODSIn this study,cerebral ischemia-reperfusion injury model was made by 2 hours of middle cerebral artery occlusion(MCAO) followed by 24 hours of reperfusion in rats′right cerebral.There were 8 rats alive every group after making model.Rats were pretreated with puerarin 25,50,100 mg·kg-1,nimodipine 0.7 mg·kg-1 once a day for 7 days and then subjected to MCAO.We examined neurological infarction sizes by 2,3,5-triphenyltetrazolium chloride(TTC) staining technique,the cerebral water contents measuring by wet/dry weighing method,the neurological deficit scores grading on a scale of 0–4.The effects of puerarin at the dosage of 25,50 and 100 mg·kg-1 were compared respectively with nimodipine 0.7 mg·kg-1 and model group.RESULTS Puerarin 50 and 100 mg·kg-1 could obviously decrease infarction volumes,neurological deficit scores and cerebral water contents after MACO(P 0.05 or P0.01 vs.model group).CONCLUSION Puerarin could protect against the cerebral ischemia-reperfusion injury in rats.
