Title: Expression of survivin protein in human non-small cell lung cancer and its significance between survivin and caspase-3, p21~ WAF1 proteins
Abstract: OBJECTIVE:To investigate the expressions of survivin, caspase-3 and p21~ WAF1 proteins in non-small cell lung cancer (NSCLC) and their relationship between their expressions and clinical pathology parameters, and the correlation between survivin protein expression and caspase-3, p21~ WAF1 protein expressions respectively. METHODS: The serial sections of the same tissues were subjected to immunohistochemical stain of survivin, caspase-3 and p21~ WAF1 proteins in 87 patients with NSCLC (42 patients with squamous cell carcinoma, 45 with adenocarcinoma).RESULTS: In 87 NSCLC, survivin, caspase-3 and p21~ WAF1 protein positive rates were 71.26%, 64.37% and 48.28% respectively. The positive expressions of survivin, caspase-3 and p21~ WAF1 proteins were not associated with histology types (P0.05), but were highly associated with lymph node metastasis (P0.05); the expressions of survivin and caspase-3 proteins were not associated with cellular differentiation ( P0.05), but p21~ WAF1 protein was closely associated with cellular differentiation (P=0.003). In 87 NSCLC, a negative correlation was present between survivin and caspase-3 protein expressions (P=0.001), survivin and p21~ WAF1 protein expressions (P=0.000).CONCLUSIONS: There is a significant correlation between survivin, caspase-3 and p21~ WAF1 genes and the infiltration depth, stage in NSCLC. Survivin blocks cell apoptosis by inhibiting caspase-3 activity or shortens cell cycle by inhibiting p21~ WAF1 gene expression, thereby promots proliferation of cancerous cell of NSCLC.
