Title: Effects of two cooling modes of low temperature stress on antioxidant enzyme activities and malondiadehyde level in sea cucumber Apostichopus japonicus
Abstract: Effects of slow down and sharp drop of low temperature on activities of antioxidant enzymes catalase( CAT) and superoxide dismutase( SOD) and malondiadehyde( MDA) level were studied in sea cucumber Apostichopus japonicus with body weight of( 107. 48 ±23. 28) g. The results showed that the activities CAT and SOD and the content of MDA were shown to be increased in the fluid of the sea cucumber exposed to slow temperature down.In the 2 week recovery period,however,the activity of CAT was increased to a peak of 29. 745U/mL,and the activity of SOD recovery to the level before the low temperature stress. The activity of CAT was increased significantly to 31. 165U/mL( P0. 05) in the sharp drop low temperature stress,and the activity of SOD decreased to the minimal value of 0. 401U/mL,with increase in the content of MDA. In the 2 week recovery period,the activity of CAT was increased to a peak of 35. 154U/mL,and the activity of SOD reached to the level before the low temperature stress. There were significantly lower content of MDA and activities of CAT,and SOD in low temperature treatment than in the control group in the mode of slow down low temperature. However,there were significantly lower activity of CAT and MDA content of and significantly higher activity of SOD in the mode of sharp drop of low temperature than in the control group( P0. 05). The findings indicate that the sea cucumber exposed to sharp drop low temperature stress had greater response to low-temperature and the greater ability of protecting tissues from oxidative damage,showing cold resistance compared with the sea cucumber without low temperature stress,providing a strong help with screening the strains of sea cucumber with resistance to low temperature.
