Title: Genetic Diversity of Four Geographical Populations of Macrobrachium nipponense Revealed by RAPD Analysis
Abstract: Random amplified polymorphic DNA (RAPD) technique was applied to investigate the genetic diversity among 4 geographic Macrobrachium nipponense, which were collected from Liu’an (Anhui) ,Longganhu Lake(Anhui),Huainan(Anhui) and Gaochun (Jiangsu) respectively. Under predetermined optimal reaction conditions, 16 random primers were employed to scan the genomic DNA in 30 individuals of each of 4 geographic stocks, and a total of 173 reproducible RAPD markers with a molecular of weight ranged from 250 ~ 2000 bp. The experimental data were analyzed with PopGen 1. 32. As analyzed, the proportions of polymorphic loci of Macrobrachium nipponense was 43.4%, and the Shannon index varied from 0. 177 1 to 0. 246 6 among stocks. The genetic distance revealed that the differentiation had taken place to some extend among the Macmbrachium nipponensis. The maximum genetic distance occurred between the stock of Gaochun and Longganhu populations, and the minimum between Huainan and Liu’an populations. Clustering of the stocks was analyzed by UPGMA, and the results showed the populations of Liu’an and Huainan assembled one branch first, the last did Gaochun and Longganhu.
