Abstract:Objective To investigate the expression of glioma-associated oncogene homolog 1(GLI1)and patched homolog 1(PTCH1)in human pancreatic cancer cell lines,and analyze their sequences.Methods RT-PCR was us...Objective To investigate the expression of glioma-associated oncogene homolog 1(GLI1)and patched homolog 1(PTCH1)in human pancreatic cancer cell lines,and analyze their sequences.Methods RT-PCR was used to detect the mRNA expression of GLI1 and PTCH1 in 5 pancreatic cancer cell lines and 25 normal pancreatic tissues.The products of RT-PCR were purified and cloned into pTA2 vectors,then identified by enzyme digestion and DNA sequencing.Results Both GLI1 and PTCH1 mRNA were expressed in all 5 pancreatic cancer cell lines.Yet,there were no GLI1 and PTCH1 mRNA expression in normal pancreatic tissues.Recombinant pTA2/GLI1 and pTA2/PTCH1 were successfully constructed.The results of sequencing were identical with those in GeneBank.Conclusions The activation of hedgehog signal pathway might participate the tumorigenesis of pancreatic cancer,and could be served as a potential therapeutic target.Read More
