Title: The expression and significance of TNF-α and NF-κB genes in acute alcoholic liver injury rats
Abstract: 【Objective】 To explore the expression and significance of tumor necrosis factor-α(TNF-α) and nuclear transcription factor-κB(NF-κB) in acute alcoholic liver injury rats.【Methods】 92 Wistar rats were randomly divided into model group(n =46) and control group(n =46).The groups were established by intragastric administration.56% ethanol [10 g/(kg·d)] was put into the model group and the same dose saline was put into the control group instead of ethanol three times a day for 5 days.The expressions and distribution of TNF-α and NF-κB were detected by using immunohistochemistry.【Results】 The expression of TNF-α and NF-κB were mainly observed in hepatocytes around the central vein.Compared with the rats of control group,the expression of TNF-α in liver tissue of the model group increased obviously on day 5 than that on day 3(66.67% vs.5.56%,P 0.01;83.33% vs.11.11%,P 0.01.While the expression of NF-κB genes in the model group on day 5 and day 3 was obviously higher as compared with the control group(72.22% vs.11.11%,P 0.01;88.89% vs.16.67%,P 0.01).TNF-α and NF-κB were strongly positively correlated with the liver(r =0.687,P 0.01).【Conclusion】 TNF-α plays an important role in acute alcoholic liver injury.Moreover,it may be connected with the activation of NF-κB.
