Title: STUDIES ON THE OCCURRENCE AND PREVENTION OF HYPERIIYDRIC SHOOT IN TISSUE CULTURE OF SOPHORA JAPONICA
Abstract: The factors of affecting hyperhydric shoot formation from the anther calli of Sophora japonica L. were studied. The results showed that most of the regenerated shoots produced from the short-term cultured calli and from the calli cultured on the medium supplemented with lower concentration of BA were normal shoots, whereas many hyperhydric shoots were produced from the long-term cultured calli and the calli cultured on the medium with higher concentration Of BA. After one and a half years' subculture on the MS medium supplemented with ρ(IAA) 0.2mg/L and ρ(BA) 5.0mg/L, an the buds produced from the surface of the calli were hyperhydric buds .when those buds with cant were transferred to and cultured on MS medium with different concentrations,phytohormones and growth regUlators for a month, it was found that the BA and high concentration of KT markedly promoted the formation of hyperhydric shoots; but some concentrations of NAA or 2,4-D could promote the formation of normal shoots. The observation showed that some of the normal shoots were developed from the apical and axillary meristems of hyperhydric buds, and the others produced from the calli.
