Methods (1)The effects of the thiol-oxidizing agent-diamide on cultured human umbilical vei...Objective To investigate the induction of apoptosis in vascular endothelial cells(VEC) treated with diamide. Methods (1)The effects of the thiol-oxidizing agent-diamide on cultured human umbilical vein endothelial cell(HUVEC) were observed. (2)Cell morphology examined by light and electron-microscopy, cell cycle analysed with flow cytometry. (3)In situ cell apoptosis detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL). Results (1){HUVEC} treated with diamide shows ultrastructural apoptosis characterized by cells contraction, cytoplasm and chromatin condensation, mitochondia swelling, apoptotic bodies formation. (2)Diamide not only reduced cellular percentage in S+G-2+M period of cell cycle, but also induced HUVEC apoptosis identified by apoptotic peak during flow cytometry and TUNEL(P0.05). Conclusion Dimide made HUVEC apoptosis.Read More
