Title: HPLC determination of quercetin,luteolin and apigenin in Lamiophlomis rotata(Benth.)Kudo and its capsules
Abstract: Objective:To develop an HPLC method for separation and determination of quercetin,luteolin and api- genin in Lamiophlomis rotata (Benth.)Kudo and its capsules.Methods:The C_(18)column (5μm,4.6mm×250 mm)was used with methanol- water (1mmol·L~(-1)hexadecyl-trimethylammonium bromide)(61:39,adjust pH 3.5 with phosphoric acid)as mobile phase.The flow rate was 1.0mL·min~(-1).The detection wavelength was 370 nm.Results:Quercetin,luteolin and apigenin could be well separated.The linearity ranges were 0.244-62.5mg· L~(-1)for quercetin with correlation coefficient at 0.9994,0.244-1.00×10~3mg·L~(-1)for luteolin with correlation coefficient at 0.9999 and 0.976-2.50×10~2mg·L~(-1)for apigenin with correlation coefficient at 0.9999.The RSD were all less than 5.0%.The recoveries were 100.7%,100.5%,104.1% and the RSD were 3.1%,2.5%,4.5% respectively.Conclusion:The method is sensitive,reliable and has been applied to the analysis of Lamiophlomis ro- tata and its capsules.
