Title: Effects of pH of hypothermic lung preservation solution on lung injury of immature dog during CPB
Abstract: Objective:To observe the effects of hypot he rmic lung preservation solution with different pH on the acute lung injury of im mature dogs during cardiopulmonary bypass(CPB) and to investigate the optimal pH for hypothermic lung preservation solution.Methods:36 experimental infant dogs(30 d)were random ly divided into 6 groups.Group A was CPB alone.In group B to F,pulmonary artery was perfused with lung preservation solution at pH 6.6,7.0,7.4,7.8, 8.2 re spective ly.The pulmonary artery was clamped for 60 min and then reperfused for 60 min.T he samples were collected,and MPO,MDA and NO were determined by myeloperoxid ase method,thiobarbituric acid method and nitrate reductase method respectively. The water content of lung tissue was measured and the ultrastructure of lung tis sue was observed.Results:MPO and MDA in group C,D,E F were much lower than those in gruup A (P0.05,P0.01,P0.01,P0.05 respecti vely). MPO and MDA in group E were the lowest.However,NO in group C to F were significa ntly higher than those in group A (P0.05,P0.01,P0.01,P0. 05 resp ectively).The water contents of lung in group D and E were significantly lower t han those in group A(P0.01,P0.01).At the same time,the injuries degr ee of type Ⅱ epithelial cells and endothelial cells in the lung blood vessel were dec reased in the perfusion groups.Conclusion:The hypothermic preservation solution can decrease the acute lung injury of immature dog during CPB,and the optimal pH is 7.8.
