Abstract:Objective: To investigate the mechanism of the protective effects of SZJ against apoptosis of SH-SY5Y cells induced byβ-amyloid 25-35 and SZJ/APP17 peptide were assessed by Western blot.Methods:Based ...Objective: To investigate the mechanism of the protective effects of SZJ against apoptosis of SH-SY5Y cells induced byβ-amyloid 25-35 and SZJ/APP17 peptide were assessed by Western blot.Methods:Based on the model of apoptosis of SH-SY5Y cells induced by Aβ25-35,cell viability was determined by MTT assay.The expressions of Bax,Bcl-2,Caspase-3 of SH-SY5Y cells induced by Aβμmol/L Aβ25-35.Results:25 peptide25-35(Aβ25-35)lowered the cell viability of SH-SY5Y cells,SZJ pretreatment enhanced the cell viability of SH-SY5Y cells,and there was significant difference compared with SZJ non-pretreated groups(P0.05).25μmol/L Aβ25-35 treatment up-regulated the Bax expression of SH-SY5Y cells,down-regulated the Bcl-2 expression and SZJ pretreatment down-regulated Bax expression and up-regulated Bcl-2 expression,and difference was significant compared with SZJ non-pretreated groups(P0.05).25μmol/L Aβ25-35 treatment up-regulated the Caspase-3 expression of SH-SY5Y cells,25μmol/L SZJ pretreatment down-regulated its expression,the difference was significant compared with SZJ non-pretreated groups(P0.05).Conclusion:SZJ can antagonize Aβ25-35 neurotoxicity to the SH-SY5Y cells.The mechanism of protection is likely related to a lowering in the Bax and Caspase-3 expression and a rasing in the Bcl-2 expression.Read More
