Title: Relationship of pulmonary vlara cell secretory 16-kd protein and imbalance of Th1/Th2 in asthma
Abstract: Objective To detect the relation between CC16 and T-bet,GATA-3 of patients with asthma.Methods The plasma levels of CC16,IFN-γ and IL-4 were measured by enzyme-linked immunoadsorbent assay(ELISA)and the mRNA expression of T-bet and GATA-3 in the PBMC by reverse transcription-polymerase chain reaction(RT-PCR)from 25 exacerbating asthmatic group and 33 healthy control group.Results CC16 and IFN-γ in the asthma group were [(21.96±7.31)ng/ml,(118.73±22.59)pg/ml)] lower than that in the control group [(64.88±25.27)ng/ml,(145.53±29.50)pg/ml,respectively,all P0.01)],The IL-4 in the asthma group was(425.22±4.37)pg/ml significantly higher than that in the control group(69.72±10.15)pg/ml,(P0.01).The expression of T-bet mRNA and T-bet/ GATA-3 ratio of PBMCs in asthma were significantly higher than that in the control group(all P0.01);The expression of GATA-3 mRNA was significantly lower than that in the control group(P0.01);The level of CC16 had positive correlation with T-bet mRNA expression and ratio of T-bet/GATA-3(r=0.792,r=0.761,respectively,all P0.01).There was no correlation between CC16 with the expression GATA-3 mRNA(r=-0.146,P=0.551).Conclusion The findings suggest that CC16 has close relationship with the airway inflammation and imbalance of Th1/ Th2 of asthma.
