Title: Expression of transcription factor Oct-4 in oral squamous epithelial carcinoma and its relationship with cancer stem cells
Abstract: Objective:To Investigate the expression of Oct-4 in oral squamous cell carcinoma(OSCC) tissue, demonstrate its significance in the formation of oral squamous cell cancer and relationship with cancer stem cells.Method:In 40 oral squamous cell carcinoma tissues and 3 normal mucosa tissues, the expression of Oct-4 was examined by using the streptavidin-biotin peroxidase(S-P) method.Result:In contrast to negative expression in 3 normal oral mucosa tissues, 40 of OSCC cases are all showed positive expression. There was significant difference among different clinical stages by immunohistochemistry integrate optical density (P0.01=.The rate and level of Oct-4 expression has obvious relationship with the grade of OSCC.Conclusion:The data suggest that the key transcription factor of stem cell, Oct-4 was not expressed in normal oral mucosa tissues. But it showed the expression in all the OSCC cases. This minor population of cancer cells posses the potency to induce and maintain the malignant expression of OSCC. The level of the expression of Oct-4 can be used as the biological mark of the differentiation and malignance of OSCC. This has significance applications for the final isolation and identification of OSCC cancer stem cells. The findings of our research showed the new target for cancer stem cell of OSCC and might helpful for the early diagnosis, treatment and prognosis of OSCC.
