Title: Effects of fasudil on ventricular remodeling and Rho GTPases activity in rats with heart failure
Abstract: AIM:To study the relationship between ventrieular remodeling and tissue Rho GTPases activity based on observing the effects of Rho kinase inhibitor fasudil in cardiac tissue of pressure overload heart failure model rats.METHODS:Twenty female Wistar rats with heart failure were divided randomly into 2 groups (n=10):fasudil group (fasudil 5 mg·kg~(-1),bid,ip) and model group (sodium chloride 0.1 mL, bid,ip).Ten age-matched rats received sham operation were as sham-operation group (sodium chloride 0.1 mL, bid,ip).After 4-week treatment,Rho GTPases activity was investigated in the three groups.RESULTS: Fasudil contributed to lower the ratio of left ventricular weight to body weight and relieve ventricular remodling (P0.01),and Rho GTPases activity was lowered in cardiac tissue (P0.01).CONCLUSION:Fasudil can inhibit tissue remodeling in left ventricle through lowering Rho GTPases activity.
