Title: Effects of PBDE-47 on cell cycle, apoptosis and p53 protein expression in human neuroblastoma SH-SY5Y cells.
Abstract: Objective To investigate the effects of PBDE-47 on cell cycle,apoptosis,and the protein expression levels of p53 in human neuroblastoma SH-SY5Y cells. Methods The distribution of cell cycle and the percentage of apoptosis were measured by flow cytometry,and the protein expression levels of p53 were detected by Western blot after the SH-SY5Y cells were exposed to PBDE-47 at the doses of 0, 0.5, 2.5 and 5 μg/ml respectively for 24h in vitro. Results Compared with the control group,the cell number in G0/G1 phase was significantly increased,while the cell number in S phase was significantly decreased in cells exposed to PBDE-47 (P0.001). Compared with the control group,the percentage of apoptosis was extremely increased in the cells exposed to the high dose of PBDE-47 (P0.001) and the protein expression levels of p53 in the middle and high doses PBDE-47-treated groups were significantly increased (P0.01). Conclusion PBDE-47 can induce cell cycle arrest at G0/G1 phase, increase cell apoptosis, and increase the expression of p53 protein,which indicates that p53 protein may play a role in the cell cycle arrest and cell apoptosis induced by PBDE-47.
