Abstract:Recombinant attenuated Salmonella vaccines were constructed by transforming Salmonella X4550 strain with recombinant DNA plasmids carrying Infectious bronchitis virus (IBV) structural genes S1,M and N...Recombinant attenuated Salmonella vaccines were constructed by transforming Salmonella X4550 strain with recombinant DNA plasmids carrying Infectious bronchitis virus (IBV) structural genes S1,M and N (pVAX1-S1,pVAX1-M,pVAX1-N). Expression of recombinant proteins in vitro was confirmed by immunofluorescence assay. These Salmonella vaccines were administered to SPF chickens via oral route and their distributions and stability in vivo was monitored. IBV-specific IgG antibody,mucosal IgA antibody,and the number of CD4+,CD8+ T cells were detected. Results showed that the recombinant IBV attenuated Salmonella vaccines could be detected in heart,liver,spleen,lung,kidney and remained stable in these organs. IBV-specific IgG antibody,mucosal IgA antibody and the number of CD4+,CD8+ T cells increased significantly following vaccination. Challenge with virulent IBV M41 strain showed that 73% vaccinated chickens was protected. This method highlighted the feasibility of developing alternative IBV vaccines using Salmonella as a carrier.Read More
