Title: APPLICATION OF PAT FOR IN-LINE MONITORING OF POWDER MIXING PROCESSES AND CHEMICAL IMAGING OF PHARMACEUTICAL FORMULATIONS
Abstract: Driven by the Process Analytical Technology (PAT) initiative, spectroscopic techniques like near-infrared (NIR) and Raman spectroscopy are implemented for online monitoring and process control. Here, we present results on quantitative in-line and real-time monitoring of powder mixing processes by near-infrared (NIR) spectroscopy [1]. Spectral data were collected with an FT-NIR Spectrometer and data analysis was performed with TheUnscrambler (Camo) and SIMCA-P+ (Umetrics) following different strategies with single and multiple probe setups for determination of the mixing end-point and for a spatially resolved monitoring of the powder flow kinetics within a blender. In addition to NIR spectroscopy, we introduce a hyper spectral imaging system (EVK DI KERSCHHAGGL GmbH) to monitor fast continuous processes. For noninvasive product quality analysis of solid dosage forms, spectral Raman mapping is applied to investigate coating homogeneity and the
