Title: Isolation and identification of porcine reproductive and respiratory syndrome virus NM1 strain and sequence analysis of its Nsp2 gene.
Abstract: A porcine reproductive and respiratory syndrome virus(PRRSV) field strain,PRRSV NM1,was isolated from a sick pig with high fever on a pig farm in Inner Mongolia,China.The strain replicated well in Marc-145 cells and caused cytopathic effect(CPE) on cell culture.PRRSV NM1 strain was identified to be a member of America genotype based on RTPCR analysis.The Nsp2 gene of the strain was amplified and sequenced.The results revealed discontinuous deletions of 30 amino acids in the peptide deduced from the Nsp2 gene.Sequence analysis showed that the Nsp2 gene shared 99.1%,99.0% and 98.9% nucleotide homologies compared with other high pathogenic PRRSV strains HuN4,HuB2 and JXA1,respectively.Five 25-day-old PRRSV-free piglets were inoculated intranasally with PRRSV NM1 strain by virulence test.All piglets were infected with the strain and 3 of them died in the test.It suggested that PRRSV NM1 strain was a variant strain from other strains,having a strong pathogenicity.
