Title: A fluorometric protein assay suitable for histones based on lysine content
Abstract: A new, simple, and highly sensitive method for measurement of histones is based on fluorescence after treatment with fluorodinitrobenzene. The fluorescence is related to the lysine content of the protein. The data indicate that DNP-lysyl residues in proteins are much more fluorescent than free DNP-lysine. It is posiible to detect less than 0.2 μg of lysine-rich histone or 0.4 μg arginine-rich histone by this method. Most of the commonly employed reagents for histone extraction or fractionation do not interfere. The method is valuable for monitoring chromatography of small amounts of histone, and when used in conjunction with other physical measurements permits a distinction to be made between histones and other proteins, and berween lysine-rich and arginine-rich histones.
