The hydroxylysine: hydroxyproline ratio in basement membrane was 0.42 compared with 0.07 in collagen. Carbohydrate makes up approximately 10% of basement membrane by weight, while collagen contains 0.6% carbohydrate. Basement membrane was solubilized by reduction of disulfide bonds in 8 M urea followed by alkylation.
