Title: Genetic diversity of three wild Exopalaemon modestus populations from Jiangsu Province revealed by AFLP markers.
Abstract: The genetic diversity and genetic structure of three wild populations of the freshwater prawn Exopalaemon modestus in Jiangsu Province were investigated using AFLP markers.A total of 102 unambiguous polymorphic bands were detected with three primers combinations(E-AAC/M-CAA,E-AAC/M-CAG,E-AAC/M-CAT),of which 93 were polymorphic loci.A fairly high genetic diversity was found at the level of species and among the populations.At the level of species,the percentage of polymorphic loci was 91.18%,Nei's gene diversity index(h) was 0.391 and Shannon information index(I) was 0.562.The percentage of polymorphic loci of 3 populations ranged from 58.82% to 70.59%,with average of 65.4%.The population with the highest percentage(70.6%) of polymorphic loci was found in the Luomahu Lake,and the population with the lowest percentage polymorphism(33.64%) was the one collected from Yangtze River in Nanjing.Nei's gene diversity of the 3 populations was from 0.268 to 0.320,with average of 0.292.The Shannon information index was from 0.381 to 0.454,with average of 0.417.The genetic differentiation between the population of Yangtze River in Nanjing and the population of Tai Lake was lower than other pair of populations.It was suggested that effective measures should be taken to avoid the degeneration of source germplasm in the reproduction and breeding of the prawn.
