Title: Isolation and characterization of Sclerotium rolfsii causing leek blight.
Abstract: Effects of temperature, pH, light,moisture , nutrient and fungicides on growth of Sclerotium rolfsii Sacc. were investigated. The range of temperature for the mycelium growth was 13-40 ℃, the optimum at 31 ℃. The range of pH was 2-9, the optimum at pH 6. Red light was the best light to it's growth. The best carbon source was sorbierte and sucrose. The fungus could not survive on lactose. Among the 9 nitrogen sources tested, peptone gave the best growth. The range of temperature for sclerotia germination was 16-40 ℃,the optimum at 34 ℃.The range of pH was 2-10, the optimum at pH 3-4.Under RH 100 %+water, the sclerotia could germinate well. Green light was the best light to it's germination. In addition, light was good for sclerotia production. White light was the best light to the sclerotia production.Among the 11 fungicides tested, these fungicides, such as propiconazole, myclobutanil, triadimefon, difenoconazole and prochloraz, gave a strong inhibition to the growth of the fungus.
