Title: The Architecture of Photosystem II in Plant Photosynthesis Which Peptide Subunits Carry the Reaction Center of PS II?
Abstract: Photosystem II in the thylakoid membrane of plant chloroplasts catalyzes oxygen evolution and plastoquinone reduction. Enriched photosystem II particles contain a reaction center P680, pheophytin, Fe, plastoquinones QA and QB, Mn (for the oxygen-evolving system), core antenna chlorophyll a and a cyrochrome b559 of unknown function. The protein complex consists of 47, 44, 34, 32 and 10 kD peptide subunits, probably in ratios of 1 : 1. Experiments will be presented with trypsinated photosystem II particles, still capable of oxygen evolution and QA reduction, that do not support the notion in the literature that the 47 kD peptide is the subunit carrying the reaction center. Instead it is suggested that the reaction center of PS II consists of a complex of the 34 and 32 kD peptide. The amino acid sequence and particularly their predicted membrane folding is highly homologous to the L and M subunit of the bacterial reaction center. It is therefore very suggestive that the principle architecture of photosystem II is quite comparable to the photosystem of purple bacteria.
