Title: Unaltered D-[3H]mannoheptulose uptake after prior in vitro exposure of rat pancreatic islets to streptozotocin
Abstract: This study aims to investigate whether the prior exposure of rat pancreatic islets to streptozotocin (3.8mM) for 60min at 37°C alters the uptake of D-[ 3 H]mannoheptulose (0.1mM) in islets that were cultured for 24 hours after the exposure to the β-cytotoxic agent and then incubated for 60min in the presence of the tritiated heptose. Under these conditions, no significant difference between control and streptozotocin-affected islets could be detected in terms of the apparent intracellular distribution of D-[ 3 H]mannoheptulose. This situation contrasts with that found when comparing the uptake of D-[ 3 H]mannoheptulose by pieces of pancreas from control animals versus streptozotocin-induced diabetic rats, indicating that the impaired uptake of the tritiated heptose by the pancreas of the latter rats is attributable to the decrease in B-cell mass.
