Title: WORKSHOP SUMMARY: ENZYMOLOGY OF BASE EXCISION REPAIR
Abstract: The base-excision repair is a major component for maintaining DNA integrity. Evidence has been presented for the involvement in E. coli of uracil-DNA glycosylase and the AP endonuclease activity associated with exonuclease III (endonuclease VI) in the removal of uracil from DNA. The use of apurinic DNA as substrate yields a different enzyme from that obtained with alkylated DNA substrates. There are evidences for the involvement of uracil-DNA glycosylase in uracil excision. Uracil-, hypoxanthine-, and 3-methyladenine-DNA glycosylases are evidently not physically associated with the enzymes studied in Goldthwait's laboratory that can release O6-methylguanine and bromomethyl-benzanthracene from DNA. Once the spectrum of these enzymes is established in bacteria and in eukaryotic cells, the base-excision repair is found to be more important than initially anticipated.
