Title: Los ácaros Laelapidae (Parasitiformes, Mesostigmata) parásitos de los roedores de la tribu Phyllotini (Cricetidae, Sigmodontinae): sistemática y relaciones filogenéticas
Abstract: La historia evolutiva de los hospedadores influye en sus comunidades de parásitos. Cuando los hospedadores son similares o cercanos filogenéticamente, debido a la coevolución y coespeciación, las comunidades parasitarias asociadas tenderán a ser compartidas. Aunque esto puede atenuarse, debido a procesos de extinción, “missing the boat”, “host-switching” o cuando los parásitos son extremadamente generalistas. Los lelápinos constituyen un ejemplo de asociación a roedores cricétidos sigmodontinos. En muchos de sus casos, las asociaciones son muy estrechas con sus hospedadores. En Argentina, tanto los lelápinos como sus hospedadores sigmodontinos tienen una gran representación, destacando a la tribu Phyllotini, por ser una de las más diversas. Phyllotini se divide en dos subtribus: Calomyina y Phyllotina. Su diversidad, amplitud territorial e importancia zoonótica la convierten en un modelo ideal para evaluar su comunidad de lelápinos. Además, en Argentina se presentan tres regiones biogeográficas, con condiciones climáticas contrastantes, lo que permitirá evaluar si estas comunidades también responden a un componente geográfico. El objetivo principal de esta tesis fue estudiar la comunidad de ácaros Laelapinae (Mesostigmata: Laelapidae) parásitos de roedores de la tribu Phyllotini (Cricetidae: Sigmodontinae) con el fin de explorar su taxonomía y poner a prueba si estos reflejan la estructura filogenética de sus hospedadores. Para esto se estudiaron ácaros lelápinos obtenidos de filotinos de 44 localidades de Argentina, representados por los géneros Calomys (6 spp.), Eligmodontia (2 spp.), Graomys (2 spp.), Loxodontomys (1 spp.), Phyllotis (3 spp.) y Tapecomys (1 spp.). Este material se complementó con otro conseguido en préstamo de otras localidades de Argentina, Chile y Perú, representados por los géneros Calomys (4 spp.), Graomys (1 spp.), Loxodontomys (2 spp.) y Phyllotis (3 spp.). Este material complementario fue utilizado únicamente para comparaciones morfométricas y genéticas. A una submuestra de ácaros se les realizó extracción de ADN, mediante un método no destructivo, posteriormente se realizó una PCR para las regiones nucleares del espaciador transcripto interno 1 y 2 (18S-ITS1-5.8S-ITS2-28S) y del citocromo oxidasa 1 (COI). Después de la extracción de ADN, se recuperó la cutícula del ácaro, la cual fue aclarada y montada para su observación mediante microscopio óptico. En este proceso, los ácaros fueron identificados, medidos y fotografiados. Se realizaron distintos análisis para morfometría lineal y geométrica, para evaluar la variabilidad de las medidas y las formas de los ácaros entre las distintas especies de hospedadores. Luego se compararon las secuencias de ADN obtenidas con secuencias de lelápinos previamente disponibles en Genbank. De los 398 filotinos analizados se obtuvieron 1584 lelápinos, el 85% correspondió Laelaps mazzai (Fonseca, 1939) y el 15% restante a Androlaelaps fahrenholzi (Berlese, 1911). Se observó que L. mazzai es una especie central en Calomyina, mientras que A. fahrenholzi es una especie secundaria. Por otro lado, Phyllotina estuvo exclusivamente parasitado con A. fahrenholzi. Hubo mayor diversidad de ácaros en la región Neotropical, con respecto a la región Andina. Los análisis morfométricos indican que L. mazzai y A. fahrenholzi son ácaros polimórficos y que tienden a mostrar diferencias con respecto a sus hospedadores. Los análisis moleculares de L. mazzai muestran que existe alto flujo génico entre poblaciones de este ácaro, sin embargo, en A. fahrenholzi la estructura filogenética muestra dos clados, cada uno asociado a las regiones biogeográficas Neotropical y Andina, mientran que en la zona Transicional Sudamericana se superponen las secuencias. Se describió por primera vez la deutoninfa de L. mazzai, reconocida, entre otras características, por la marcada hipertriquia en el idiosoma. Los resultados de esta tesis son congruentes con una historia coevolutiva estrecha entre L. mazzai y Calomyina. Por su parte, Laelaps mostró una historia de coevolución con los roedores Murinae y Sigmodontinae. Androlaelaps fahrenholzi mostró una filogenia más asociada al ambiente abiótico; sin embargo, podría tener alguna diferenciación específica en roedores del género Loxodontomys. Las investigaciones de esta tesis revelan que la comunidad de ácaros lelápinos en roedores filotinos está compuesta por A. fahrenholzi y L. mazzai. Mientras que la primera es altamente generalista, L. mazzai tiene un nivel de especificidad con Calomys. La composición de la comunidad es característica de cada subtribu de filotinos (Calomyina y Phyllotina) y se mantiene a lo largo de su distribución geográfica, con escasas excepciones. Tanto la presencia de L. mazzai en Calomyina, como su ausencia en Phyllotina, pueden deberse a procesos coevolutivos. Laelaps mazzai muestra una alta preferencia por Calomys, probablemente debido a una adaptación a climas áridos. Se identificaron nuevas especies hospedadoras y zonas geográficas para L. mazzai y A. fahrenholzi, ampliando sus registros hacia el sur de Sudamérica y en Perú (en el caso de L. mazzai). Las variaciones morfológicas y genéticas de los lelápinos asociados a filotinos no son suficientes para proponer nuevas especies, concluyendo que A. fahrenholzi y L. mazzai son especies polimórficas. Futuros estudios con nuevos marcadores moleculares y más muestreos resultan necesarios para completar la historia filogenética de estos ácaros en Sudamérica. Las especies de Laelaps muestran una divergencia profunda entre Murinae y Sigmodontinae, aspecto que sugiere implicaciones taxonómicas y nomenclatoriales relevantes.
