Title: Chapter 2 History of ultrasensitive enzyme immunoassay
Abstract: Publisher SummaryThis chapter provides the history of ultrasensitive enzyme immunoassay. Radioimmunoassay, developed in 1959, made it possible to measure femtomole amounts of peptide hormones in the circulation. Later, the use of enzymes as labels in an immunoassay was suggested, and the first attempts of an enzyme immunoassay were reported in 1971. For many years later, the potential of an enzyme immunoassay, particularly concerning its sensitivity, was a focus of discussion by many investigators. For antibodies and haptens, both the assay system and the sensitivity of an enzyme immunoassay were fundamentally unchanged for many years after their first reports. In 1987, an ultrasensitive enzyme immunoassay for antibodies was developed. Recently, the period required for ultrasensitive enzyme immunoassays has been considerably shortened with further improvement of the sensitivity not only to antigens but also to antibodies.
