Title: Synthesis of tobacco mosaic virus infectivity by cell-free extracts
Abstract: An improved and simple method is described for preparing extracts from TMV-infected leaves which engender a rise in TMV infectivity simultaneously with incorporation of [14C]ATP into a material having the properties of RNA. The rise in infectivity depends on the presence of ATP, GTP, CTP and UTP in the incubation mixture. Incorporation occurs with ATP alone, but to a greater extent when the four triphosphate nucleotides are present. 2. The incorporation and rise in TMV infectivity reactions are time dependent, inactivated by ribonuclease (EC 2.7.7.16), and heat.
