{'薬物混入飼料(DAF)法によりmorphineを適用したマウスに,morphineの反復投与を行って,morphineの適用条件とmorphineの反復投与による自発運動促進作用変化について検討した.実験にはICR系マウスを用い,自発運動の測定はtilting法により行った.普通飼料を与えたマウスでは,morphine': [0], '10mg/kgを3~4日に1度の割合で反復皮下投与すると,自発運動促進作用の増強すなわち逆耐性現象が観察された.これに対して,morphine混入飼料1mg/g': [1], 'foodを適用した条件で,同様にmorphineを反復皮下投与しても,自発運動促進作用の増強は観察されなかった.したがって,DAF法による適用のように,少量であっても常にmorphineに動物が暴露されている適用条件では,この自発運動促進作用の増強(逆耐性)は生じないものと考えられる.さらに,morphine混入飼料を1~3mg/g': [2], 'foodに漸増適用する条件では同様にmorphineの反復投与を行っても自発運動促進作用の増強はみられず,むしろ自発運動促進作用の抑制が観察された.また,morphine混入飼料を漸増適用したマウスにmorphineを単回皮下投与した場合においても,同様に自発運動促進作用の抑制が観察された.このmorphine混入飼料の漸増適用は,マウスに強い身体依存を形成する適用方法である.一般に,morphineの中枢抑制作用には耐性が生ずるが,中枢興奮作用に対しては耐性が起こらず,逆耐性が生じると報告されている.したがって,DAF法によるmorphineの漸増適用のように,morphineをマウスに連続的に適用し,身体依存を形成させるような適用条件下では,morphineの中枢興奮作用である自発運動促進作用にも,耐性が生じることが示唆された.':
