Pharmacol. 4 (1968) 228–230The uptake of 3H-ouabain and 3H-digitoxin was studied in guinea-pig atria that had been pretreated with 50% aqueous glycerol for 8–10 days. At equilibrium, the tissue/medium radioactivity ratios for 3H-ouabain and for 3H-digitoxin were 0.7 and 4.7, respectively. Whereas the uptake of 3H-ouabain by the glycerol-treated organs was the same as that by normal, beating atria, the relative accumulation of 3H-digitoxin was considerably reduced. The results suggest that in atrial tissue digitoxin is present in both lipid and protein containing cellular compartments, whereas ouabain is mainly dissolved in the aqueous constituents of the cells and hardly any of the drug is bound to cellular structures.
