Title: Cell-free synthetic biology as an emerging biotechnology
Abstract: Over the past few decades, the field of synthetic biology has transformed and will continue to advance biotechnology. However, synthetic biology tools have relied on living cells, thus limiting the application and scope of synthetic biology. Cell-free protein synthesis or gene expression systems enable open, modular, and flexible environments for engineering biological systems by minimizing complexity and maximizing programmability, thus providing unprecedented freedom in the development and application of synthetic biology tools. Herein, we summarize how cell-free systems have accelerated synthetic biology to produce difficult-to-express functional proteins and biomaterials, use genetic circuits for exploring biological networks, develop novel biosensors and diagnostics, create artificial cells, and improve metabolic engineering by enabling rapid prototyping and novel biosynthetic pathway creation capabilities. Cell-free synthetic biology should play an increasingly crucial role in establishing affordable and efficient biological platforms, and producing many innovative outcomes.
