Title: Efficiency the hairy roots of radish ( Raphanus Sativus ) plant which genetic transformed by Agrobacterium Rhizogenes ATCC15834 for anthocyanin production
Abstract: The present study succeeded in initiated the genetically transformed hairy roots from Raphanus sativus L. seedlings leaves as a result of inoculated with Agrobacterium rhizogenes ATCC15834 suspension at 24,48,72 hours after incubation, the genetic transformation ratio were 40, 66.66, 0 % respectively after 20 days. The results indicated the development of those roots when cut and placed on the solid MSO medium to prduced well-grown cultures and the hairy roots characteristic as white in color and grow in many branches with added Cefotaxime antibiotic to the medium when it contaminated with bacteria. The study was able to improve the genetic transformation at molecular level by isolated the DNA from hairy root samples of both types and amplifying it with Polymerase chain reaction (PCR) technology. After the electrophoresis, the results revealed that there is one band for each type of DNA with a molecular weight at 248bp similar to the molecular weight of the specialized primer of the rol A gene. The quantities of anthocyanins recorded a superiority in their concentrations determinate in the genetically modified hairy root samples than in the radish seedlings root samples.
