Title: Optimization of ultrasonic-assisted extracting polysaccharide from banana leaves
Abstract: 【Objective】The ultrasonic-assisted extraction process of polysaccharide from banana leaves was optimized to provide theoretical references for further development of banana leaves. 【Method】Banana leaves without monosaccharide after degrease were used as research materials. On the basis of single factor experiments, a three-factor and three-level Box-Behnken central composite design was utilized for mathematical modeling of polysaccharide yield. Influencing factors included extraction temperature, extraction time and ultrasonic power. With polysaccharide extraction rate as response value, response surface analysis was made to optimize extraction conditions. 【Result】 Polysaccharide content in banana leaves on the coding independent variables were extraction temperature, extraction time and ultrasonic power and the quadratic multinomial regression equation was Y=1.42+0.064A+0.065B+0.00775C-0.057A2-0.057B2+0.003525C2+0.022AB-0.0075AC+0.006BC. The optimum conditions for the extraction of polysaccharide from banana leaves were as follows: extraction temperature of 66.35 ℃, extraction time of 28.73 min and ultrasonic power of 150 W. The polysaccharide yield of the experiment was 1.40% under the conditions of extraction temperature of 66 ℃, extraction time of 29 min and ultrasonic power of 150 W. 【Conclusion】Ultrasonic-assisted extraction of polysaccharide from banana leaves could decrease extraction temperature and greatly reduce the extraction time. So the extraction method was efficient.
