Abstract: The physical requirements that are optimum for bacterial growth vary dramatically for different bacterial types. Microorganisms can be divided to five groups according to the oxygen requirements: aerobic, facultative anaerobic, microaerophilic, anaerobic microorganisms, and aerotolerant anaerobic (fermentative). Oxygen tension is always higher near the surface area of the medium. Medium will be colored at the top because of presence of O2, and colorless in the middle and bottom because of absence of free O2. The medium containing lower amount of agar restrict the microorganisms to small area and provides anaerobiosis through medium in tube. In this practice, the effects of O2 on the growth of microorganisms will be indicated.
