Abstract:Objective To investigate the expression and significance of homeobox protein EN2 in human prostate cancer (PCa).Methods EN2 expression in paraffin embedded tissue of human prostate cancer (study group...Objective To investigate the expression and significance of homeobox protein EN2 in human prostate cancer (PCa).Methods EN2 expression in paraffin embedded tissue of human prostate cancer (study group,n =48 ),High grade prostatic intraepithelial neoplasia (HGPIN,the first control group,n =22) and benign prostatic hyperplasia (BPH,the second control group,n =48) were investigated by immunohistochemical.EN2 expression in fresh tissue of human prostate cancer (study group,n =14 ) and BPH ( control group,n =15 ) were determined by Western blotting.Results The expression level of EN2 in prostate cancer tissue was higher than those in the HGPIN and BPH tissue by immunohistochemical.EN2 protein was presented in the cytoplasm mainly and in some cases in the nucleus.The positive rate in PCa,HGPIN and BPH were 87.50%,31.81% and 27.08% respectively.The difference was significant statistically ( P < 0.0l ).The positive rate in well-differentiated PCa ( n =4),moderately-differentiated PCa ( n =17 ) and poorly-differentiated PCa ( n =27 ) tissue was 75.00%,88.24% and 88.90% respectively.The results of Western blotting showed the expression level of EN2 in prostate cancer was 3fold higher compare to BPH.Conclusion Homeobox protein EN2 was expressed ectopically in prostate cancer and secreted by prostate pithelial cell.
