Title: Effects of sevoflurane postconditioning against ischemia injury and on the expression of insulin-like growth factor-1 in rat hippocampus
Abstract: Objective To observe effects of sevoflurane postconditioning against ischemia injury and on the expression of insulin-like growth factor-1 in rat hippocampus with different inhalation doses. Methods Experiments were performed using male SD rats which were randomly assigned to 6 groups: sham operation group (group S); ischemia-reperfusion group (group C); oxygen postconditioning group (group O_2); sevoflurane postconditioning group with inhaled concentration of 1.5% (group Sevol), 2.4% (group Sevo2) or 3.0% (group Sevo3). All animals were subjected to the modified model of right middle cerebral artery occlusion (MCAO) for 120 min. The histopathology of hippocampal slices and the IGF-1 mRNA expression of hippocampus were recorded at 24, 48, and 72h after reperfusion. Results In group C, IGF-1 mRNA expression increased and the obvious histopathologic damage in hippocampus was observed. The IGF -1 mRNA expression further increased and the histopathologic damage in hippocampus attenuated in groups Sevo2 and Sevo3, but not in group Sevol, compared to that in group C. Conclusion Sevoflurane postconditioning with higher inhalation dose exert neuroprotective effects against ischemia injury via up-regulating IGF-1 receptor system in rat hippocampus.
